Extra details

Available ordering format:Lyophilized in PBS pH 7.2
Immunogen:ascorbate oxidase purified from Cucurbita sp.
Raised in:Rabbit
Purification:Total IgG
How to reconstitute:For reconstitution add 1 ml of sterile water
Storage condition:store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please, remember to spin tubes briefly prior to opening them to avoid any losses that might occur from lyophilized material adhering to the cap or sides of the tubes. Working dilutions should be stored at 4°C, not refrozen and prefarably used the same day. If slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the product.
Verified applications:Western blot (WB), ELISA (ELISA)
Connected products:AS06 184 | anti-ascorbate oxidase rabbit antibodyPlant and algal protein extraction bufferSecondary antibodies
Recommended dilutions for use:1: 1000 (WB), 1: 1000 - 1: 30 000 (ELISA)
Molecular weight (expected | аpparent):61 | 45 kDa (Cucurbita pepo)
Verified reactivity:Cucurbita sp.
Possible reactivity:dicots including Glycine max, monocots including Oryza sativa
No reactivity:no confirmed exceptions from predicted reactivity known in the moment
Supplementary information:IgG concentration is 10 mg/ml. Biotin/IgG protein molar ratio is approximately 5.6. No foreign proteins are added.
References:to be added when available
Scientific context:Ascorbate oxidase (AO) is an apoplastic enzyme involved in metabolism of plant ascorbate (AA). Ascorbate (AA) plays a key role in defense against oxidative stress and is particularly abundant in photosynthetic tissues. Over 90% of the ascorbate is localized in the cytoplasm, but a substantial proportion is exported to the apoplast.
Notes:IgG protein fraction is prepared by ammonium sulphate precipitation and ion exchanged chromatography. N-Hydroxysuccinimidobiotin is used for labelling of antibody.
Protein number:Refer to NCBI
TAIR number:Refer to NCBI
Properties:Biotin conjugates can be detected by horseradish peroxidase, alkaline phosphatase substrates or anti biotin conjugated antibodies. Avidin and Streptavidin bind to the small biotin and are couple to HRP or AP for ELISA. To break the streptavidin Biotin bond we suggest to use a 6 molar guanidine HCl solution with acidity of pH 1.6.
